چکیده

The aim of this study was to investigate the effects of insulin therapy on glucagon in patients with newly diagnosed Type 2 Diabetes (nd-T2DM patients). We recruited 93 nd-T2DM patients, including 45 nonobese patients and 48 obese patients. A 100 g bread meal test was performed before and after insulin therapy, and glucagon levels were measured before and after the experiment. Compared with the control group, the serum glucagon levels before treatment in both the obese and non-obese nd-T2DM patients were significantly higher (P=0.001). After treatment, the serum glucagon levels in the nd-T2DM patients had significantly decreased (P=0.001), but they were still higher than those in the control group (P=0.001). Additionally, the area under the curve of serum glucagon and postprandial glucagon levels in the non-obese nd-T2DM patients decreased significantly (P=0.001 and P<0.01, respectively). The serum glucagon level in the obese nd-T2DM patients decreased non-significantly (P>0.05). Insulin therapy improved serum glucagon levels in nd-T2DM patients. The serum glucagon level in the non-obese ndT2DM patients improved significantly, but that in the obese nd-T2DM patients did not. The CP levels in the non-obese T2DM group at 30 min (P=0.003), 60 min (P=0.001), 120 min (P=0.001), and 180 min (P=0.001) after treatment had significantly increased compared to that before treatment. The CP levels in the obese T2DM group at 120 min (P=0.001) and 180 min (P=0.001) after treatment had significantly increased compared to that before treatment. This improvement might be related with a potential association between T2DM and obesity.

Effects of sitagliptin as initial therapy in newly diagnosed elderly type 2 diabetics: A randomized controlled study

The objective of this study was to investigate the characteristics of blood glycemic excursion, incretins and pancreatic hormone secretion in elderly people with newly diagnosed type 2 diabetes mellitus (T2DM) and to study the effects of sitagliptin on glycemic excursion in these subjects. A total of 129 newly diagnosed T2DM patients were enrolled in the study from March 2012 to August 2013. Cl...

Unusually rapid β-cell failure in a patient newly diagnosed with type 2 diabetes presenting acutely with unprovoked severe hyperglycaemic hyperosmolar state: a case report

Pancreatic beta-cell failure on a background of insulin resistance results in the inability to compensate for fasting hyperglycaemia and eventually produces type 2 diabetes mellitus. We describe an interesting case of a patient who presented acutely with unprovoked severe hyperglycaemic hyperosmolar state and was subsequently diagnosed with type 2 diabetes mellitus on a background of only impai...

Serum Glucagon-Like Peptide-1 Changes in Women with Type 2 Diabetes Following a Four Weeks Aerobic Exercise

Objective: The objective of this study was to investigate the effect of four weeks aerobic training on serum glucagon-like peptide-1 (GLP-1) in women with type 2 diabetes mellitus (T2DM). Materials and Methods: Twenty T2DM patients (33-53 years) were randomly assigned into control (n=10) and experimental (n=10) groups. The main intervention included running on treadmill, reaching 55 to 80% o...

Single-dose acarbose decreased glucose-dependent insulinotropic peptide and glucagon levels in Chinese patients with newly diagnosed type 2 diabetes mellitus after a mixed meal

BACKGROUND Acarbose slows down the intestinal absorption of carbohydrates, but its effects on the secretion of incretins are still poorly known. This study aimed to examine the effects of single-dose acarbose on the secretion of incretins in patients with newly diagnosed type 2 diabetes mellitus (T2DM). METHODS In this pilot study, twenty-three patients diagnosed with T2DM were randomly assig...

Improvement in insulin sensitivity following intensive insulin therapy and association of glucagon with long-term diabetes remission

Objective To investigate the role of the acute glucagon response in the long-term remission of newly diagnosed type 2 diabetes mellitus following short-term intensive insulin therapy (IIT). Methods Ten patients with newly diagnosed type 2 diabetes mellitus received IIT. Intravenous glucose tolerance tests and the clamp technique were performed pre- and post-IIT. Remission was defined as mainten...
